Insulin is a peptide hormone synthesized by the beta cells of the pancreas. It has the function to facilitate the entry of glucose inside the cells to maintain normal blood glucose levels. An increased level of insulin causes insulin resistance in which cells have an impaired sensitivity to insulin causing the glucose to stay in the blood. There is increasing evidence that insulin affects the regulation of nutrient homeostasis, cognition, memory, and neuroprotection in the brain. Is insulin resistance involved in the pathogenesis of Alzheimer's disease? Definitely, in the brain, insulin resistance alongside hyperinsulinaemia promotes the formation of advanced glycation end-products, which are compounds formed by the glycation of lipids and proteins. These compounds have been suggested to increase the expression of amyloid precursor protein and amyloid beta via reactive oxygen species. Amyloid precursor protein refers to the protein from which amyloid beta plaques originate. Amyloid beta aggregation results in inflammatory response leading to the release of pro inflammatory mediators including complement factors, chemokines and eicosanoids. These pro inflammatory mediators impair microglial clearance of beta amyloid plaques and increase microglia-mediated neuronal death and damage.

In 2020, a scientific discovery was published, that could be one of the most important in the history of humanity, if confirmed by further tests currently in place.
Doctor Harold Katcher, who made this discovery, described it in the book "The illusion of knowledge", and obtained a special patent published last week.
This scientist managed, through the treatment that we will see shortly, to rejuvenate the treated rats and reduce their biological age by half, measured using one of the best clocks currently in existence.
If the possibility of reproducing this treatment in humans were confirmed, this would mean that by treating a 60-year-old individual, it would be possible to bring him back to a biological age of 30 years old.
The same scientist writes: "We have been able to reverse the aging process in rats, such that experimentally-treated old rats, measured (in terms of biological age) less than half the chronological age of their age-matched controls (untreated old animals, the same chronological age of the treated ones) according to the gold standard of biological age determination”.
He also wrote that “rejuvenation has changed from impossible dream to a new reality”. Another quote from the book is this: "my opinion has changed (and you’ll see why), and I believe that we can have biologically immortal youthfulness".
It looks like science fiction, but maybe it isn't, because there is a scientific explanation behind this rejuvenation.
Basically, from what it would seem, and trying to simplify as much as possible, rejuvenating factors, especially present in the blood of young organisms, would have been identified, factors which, if inserted into the body of an old organism, would tell the body to rejuvenate, and the body would change, rejuvenating, to adapt to these instructions given by the rejuvenating factors.
Once the treatment was done, the old rats changed, their grip strength increased considerably, the levels of the cytokines decreased towards youthful levels, and more than 30 different bio-age markers, from biochemical to higher-level cognition, showed a significant reduction in age. Memory and problem-solving seemed to have returned to near youthful levels. The levels of glutathione in the treated rats, were nearly indistinguishable from the young controls rats. Even the level of nrf2, in the brain, heart and liver, which normally decreases with age, was similar to that of young rats.
In short, our body seem to be already able to rejuvenate, but we lack ways to activate this ability, and the one just described, called E5, would be, perhaps, a way to do it.
We conclude with a last quote from the book, which makes us reflect on the ultimate consequences of this discovery. It says: “It should be possible to keep an animal at a young adult life stage indefinitely, so as load as E5 is continually provided”. He said indefinitely, in other words, forever, like the immortal jellyfish.

Doctor Shigeaki Hinohara is a medical doctor who lived to the age of 105. He was born in 1911, and died in 2017.
In the 1950s, Doctor Hinohara pioneered a system of complete annual physicals, called "human dry-dock," that has been credited with helping to lengthen the average life span of Japanese people. Women born there today can expect to live to 87; men, to 80.
The following is his diet plan for breakfast, lunch and dinner.
It is not complicated at all, since Hinohara basically ate the same thing every day.
For breakfast he drank coffee, a glass of milk and some orange juice with a tablespoon of olive oil in it.
He told The Japan Times that “Olive oil is great for the arteries and keeps my skin healthy”.
He often skip lunch.
Hinohara said that dinner was “veggies, a bit of fish and rice, and twice a week 100 grams of lean meat”.
Doctor Hinohara maintained his weight at about 130 pounds, 59 kilograms.
It is possible that his low weight contributed largely to his long lifespan, and this is a trait that many centenarians share.
It has been noted in recent books that, in the same species, short stature is often a common trait of the long-lived ones, especially when associated with a very light weight.

And it is worth noting that most centenarians are actually small and weigh very little.

Baicalein is a flavonoid extract from the roots of Chinese herb, Scutellaria baicalensis. Baicalein has been found to have anti-cancer properties in different human carcinomas. It has inhibitory effects in angiogenesis, metastasis and inflammation in cancer. The anti-cancer properties of baicalein are mainly due to its ability to induce autophagy. Autophagy is a highly conserved evolutionary process that removes unnecessary and damaged organelles and proteins. Baicalein induces autophagy through its ability to activate AMPK pathway and inhibit mTOR or raptor complex 1 expression. Activation of AMPK pathway by baicalein further inhibits mTOR pathway. MTOR pathway is one of the most frequent mutated genes in cancer. It promotes cell proliferation and metabolism that contribute to tumor initiation and progression. Therefore, the inhibition of this pathway could prevent those mechanisms that promote cancer. Baicalein has also been shown to have anti inflammatory properties through its ability to enhance the activation of NRF2 and downregulate the activation of NLRP3 inflammasome and NF kappa B phosphorylation. Baicalein and its metabolite baicalin have been reported to ameliorate lupus autoimmunity. In a study conducted in 2019 by the group of Dr. Ji Yang, 4 weeks of treatment with baicalin has been shown to reduce 24 hour protein levels, relieved kidney inflammation and inhibit serum anti dsDNA antibody production.

David Sinclair is a scientist and professor at Harvard Medical School. He is a pioneer in the study of aging, and his research has been published in more than 200 peer-reviewed papers. His work has mainly focused on understanding the molecular mechanisms that regulate aging and age-related diseases.
Unlike other scientists, he doesn't only study anti-aging... he practices it on himself.
His drug and supplement daily routine, as he mentions in his podcast, book, and interviews, consists of the following:
1 gram of resveratrol mixed in olive oil and vinegar.
1 gram of NMN.
1-2 milligrams active ingredient of spermidine.
500 milligrams of fisetin.
500 milligrams of quercetin.

Now, the question to ask is, can this routine be improved in any way to be even more effective for longevity?
First of all, a very important thing that seems absent, is the presence of compounds aimed at fighting AGE's, Advanced Glycation End Products, which play a very important role as a driver of aging.
Advanced Glycation End Products contribute to the aging process significantly. On a molecular level, AGEs create cross-links between collagen and other proteins, which can lead to stiffening of the skin, hardening of the arteries, and even the development of arthritis. In addition, AGEs contribute to an increase in blood pressure, which can lead to heart attack or stroke. The same process that leads to stiffening of the skin also increases lines on the face and gives them a droopy appearance. These are just a few examples of how AGEs affect us on a cellular level, contributing to the aging process.
But people can fight AGEs both with specific supplements, for example Benfotiamine or Carnosine, and with lifestyle interventions.

Deprenyl, also called selegiline, is a drug that has been used to treat Parkinson's disease for over 20 years. It is also used to treat depression and other conditions.
Since it is a monoamine oxidase inhibitor, it works by increasing the amount of dopamine and norepinephrine in the brain. A study sponsored by the National Institute on Aging and published in "Current Opinion in Clinical Nutrition and Metabolic Care", found that selegiline decreased depression, anxiety, anger, hostility and stress.
Its mechanism of action is not fully understood. Some scientists believe it acts by increasing the release of brain chemicals that reduce the need to make dopamine and norepinephrine. It may also prevent the breakdown of dopamine, thereby causing an overall increase in these neurotransmitters.
Deprenyl was discovered in the early 1970s by Doctor Joseph Knoll, a Hungarian doctor who was looking for a cure for cancer at the time. He noticed that deprenyl had an unexpected side effect: it seemed to extend the lifespan of rats in his experiments.
The first longevity study demonstrating that rats treated with deprenyl lived significantly longer than their saline-treated peers was published in 1988, and corroborated in many papers.
Deprenyl has been shown to extend the lifespan of mice by up to 34%. It is not the ultimate cure for aging, but it does have the potential to increase longevity.
Deprenyl has now been used in some countries, off-label, as an anti-aging drug, for over 30 years, and it is still in clinical trials today.
The FDA has not approved deprenyl for use as an anti-aging drug, but some biohackers are already experimenting with it.

The world of science sometimes goes hand in hand with intellectual property. A very recent study aimed to investigate if the combination of several anti-aging actions can cause a cumulative effect on the extension of lifespan. Harvard Medical School and Metro International Biotech made a joint study on how MIB-626, a pharmaceutical-grade preparation of NMN is able to increase blood levels of NAD+ in middle-aged, older adults in two weeks.
There has been plenty of studies with encouraging results on NAD boosts in human cells and mice, but there are very few studies that examine the efficacy of these compounds in the human body. It might be due to the fact that there isn’t a consensus on what the safe doses of NMN are.
The participants took 2 doses of 500 milligrams of MIB-626 daily, for two weeks. The results showed that it was safe and that, after two weeks, there was a significant rise in NAD blood levels. In fact, it was doubled. One of the goals of this study is to design efficacy trials for NMN in diseases.
This study was founded by Harvard researcher Doctor David Sinclair, who’s been studying and testing NMN and NAD for years. He’s even tested NAD boosting pills with US Special Forces.
The results showed that 1,000 milligrams daily, was well tolerated by the human body. There were no serious adverse reactions. Only one of the participants had some alterations in blood levels of liver enzymes. There were no changes in the body mass index of the participants. It was tested in middle-aged and older adults who were overweight and obese but medically stable.
MIB-626 results gave evidence that it was able to triple NAD blood levels. This comes to show how NMN is metabolized after administration. It should be noted that this study was not made to evaluate efficacy but tolerance, but the results show that for incoming efficacy trials, 1,000 milligrams (or even more) of NMN should be used. Doses of less than 1,000 milligrams didn’t show any consistent raise in NAD blood levels.
Also, the results of this study agree with the resulting data of previously performed trials: it seems as though NR and NMN have no significant effects on fat, glucose, insulin, and uric acid levels. However, this study was also not long enough to study or detect changes in insulin sensitivity and glucose metabolism.
Some researchers say that NMN doesn’t get into cells, and some others have proven that there are “transporters” that carry NMN into cells. Doctor Sinclair says that all those findings are still open to debate and that, the truth might be that nobody really knows how NMN works, but that it does work.
This study will bring into light how other pharmaceutical-grade NMN products could boost NAD blood levels. If other NMN formulations are able to replicate MIB-626, we might see clinical trials of NAD boosters taking form.

The Klotho protein is related to the regulation of oxidative stress and anti-inflammatory processes. Exercise is presented as a potent stimulator of the Klotho protein, but... Who is Klotho?
In Greek mythology, Clotho is the youngest of the moirais, the goddesses of destiny. She was in charge of spinning the threads with her distaff. The strands represent human life, and that is where the gene gets its name, as it is known as a life-extending gene.
The Klotho gene, which codes for a protein of the same name, has been called the suppressor gene of aging, although interest in the control of aging is ancestral, the discovery of this gene and its protein were completely fortuitous. In 1987, Kuro-O M and collaborators found a knock-out mouse for this gene characterized by a phenotype of premature aging, hypokinesia, cognitive impairment, and sarcopenia. Since then, the study of this gene has been of great interest.
Today, three isoforms of this protein are already described, Alpha Klotho, Beta Klotho and Gamma Klotho, the first being the original and best known. Alpha Klotho is secreted mainly in the kidney, parathyroid, and choroid plexus of the brain, and can also be found, in lower concentrations, in the pancreas, ovaries, testes, and skeletal muscle.
Alpha Klotho in its circulating or secreted form has been associated with antioxidant, anti-inflammatory, and anti-proliferative effects in different tissues. In addition, it has been related to different pathologies, mainly chronic and in which the effects of inflammatory processes, oxidative stress, or cell cycle dysregulation are characteristic of the disease.
Elevated blood levels of klotho protein may help preserve kidney function. Although the very exact mechanism of action of klotho has not yet been identified, the protein has been shown to influence multiple cellular and endocrine pathways. The kidney has the highest levels of klotho expression and is probably the main source of soluble klotho, so it is not surprising that klotho levels tend to be low in patients with kidney disease.
It was found a strong association between low soluble klotho and decreased kidney function, independent of many known risk factors for decreased kidney function.
This protein is very helpful for the treatment of different conditions.
Renal Disease. Alpha Klotho is used as a powerful prognostic marker, it plays an important role in the prediction of cardiovascular events in these patients.

The Japanese Keio University had a research team that investigated and showed, for the first time, that NMN, an anti-aging intervention compound, is safe to be administered to healthy humans. This research team was led by Professor Kazuo Tsubota of the Department of Ophthalmology, Professor Hiroshi Itoh of the Department of Internal Medicine, Professor Hideyuki Okano of the Department of Physiology, and Professor Masato Yasui of the Department of Pharmacology, along with a collaboration of Washington University School of Medicine in Saint Louis.
This is because, in 2015, both universities agreed to conduct joint investigations on subjects such as longevity. The study establishes that “Recent studies in animal models have shown that the administration of NMN increases the amount of nicotinamide adenine dinucleotide (NAD+) in various organs and improves age-associated functional decline and disease conditions. However, the safety of NMN in humans has remained unclear".
Since 2016, the research team has conducted a clinical trial study to investigate the safety of single administration of NMN in ten healthy men and has confirmed that it can be safely administered and that it is effectively metabolized in the body. This study is expected to inform future treatment of age-associated diseases.
For this study, 10 healthy men aged 40 to 60 years were administered a single dose of 100, 250, or 500 mg of NMN orally. This oral administrations of NMN didn’t produce remarkable changes in blood pressure or in heart rate. Urine and blood tests, used for analyzing kidney and liver function, didn’t exceed the normal levels. Also, these single doses didn’t affect sleep quality nor did they change eye functions.
During the measurement of the number of plasma metabolites produced from NMN, the team found said plasma metabolites had increased accordingly to the dosage. From this, it was concluded that orally administered NMN can be metabolized in the human body and is safe to be used at single doses up to 500mg.

Medicine is constantly renewing itself. More and more studies and research are being carried out that lead to the advancement of techniques, medications, and procedures with the aim of improving the quality of life and even human longevity.
Among these studies are those on the effects of NMN on the human body.
A natural substance previously shown to counteract the effects of aging and improve metabolic health in mice could have relevant effects in humans, according to research conducted by a team from the Washington University School of Medicine in Saint Louis, United States.
The research, published in the journal Science, is the first randomized clinical trial to analyze the metabolic effects of the administration of the compound called nicotinamide mononucleotide (NMN) in humans.
Specifically, a small trial was conducted with postmenopausal women with prediabetes that showed how this compound "improved the ability of insulin to increase glucose uptake in skeletal muscle," notes the statement issued by the University.
This compound previously studied for aging also improved "the expression of genes that are involved in muscle structure and remodeling". However, NMN treatment did not reduce "blood glucose or blood pressure, nor did it decrease circulating markers of inflammation as observed in mice," they add.
To carry out the trial, the team of researchers administered 250 mg of this compound orally to 13 women who received it every day for a period of 10 weeks. On the other hand, a total of 12 women participants received a placebo during this same period to test the differences.
"Even though our study does show that NMN has a beneficial effect on skeletal muscle, it is still too fast to make clinical recommendations with just the results of our study as a base”, stresses lead investigator Samuel Klein, the William H Danforth Professor of Medicine and Nutritional Science and director of the Center for Human Nutrition.
Although there were some good results, researchers agreed that there will have to be more studies in order to determine the beneficial effects that NMN might have in the management or prevention of diabetes. Evaluations to further investigate NMN effects in men and women are being carried out by Imai and Klein.
Shin-ichiro Imai, professor of developmental biology and medicine and one of the principal investigators said that "This is a step toward the development of an anti-aging drug, although more research is needed to fully understand the cellular mechanisms responsible for the effects observed in skeletal muscle in people”.

Rosmarinic acid is an ester of caffeic acid that is found in rosemary and in some fern and hornwort species. Rosmarinic acid has been demonstrated to have health promoting effects through its anti inflammatory, anti diabetic, antiviral, anti oxidant and antibacterial properties. In the search for solutions to extend the years of our life, different alternatives are gaining strength. One of them is therapy with certain supplements. Some of the most promising are those that increase the levels of substances that, over the years, reduce their level in our cells, accelerating aging.
Said therapies include very complex procedures that intervene in the most intimate part of the cells, the DNA, with the risks that this entails if the process is not 100% controlled. The real panacea would be an oral treatment capable of slowing down aging. And one of the substances that are achieving the best results is a derivative of the humble vitamin B3 or niacin.
Associated with aging is a reduction in the amount of a substance called NAD+ (nicotinamide adenine dinucleotide) in the mitochondria. These small organelles are found in cells and are responsible for obtaining energy from the nutrients we get from food, combining them with oxygen.
Since it was observed that, associated with different diseases, NAD+ levels were reduced, researchers thought that a treatment capable of recovering its levels could delay or cushion the effects of aging. NAD+ is utilized by sirtuins, enzymes that get activated by fasting or calorie restriction; strategies that have proven effective in extending life in mice or monkeys and improving health in humans.
This led to the development of studies to verify if indeed, treatments with these substances are capable of slowing down the signs of aging. Research with mice in 2013, led by David Sinclair, kicked off numerous studies with rodents where improvements in muscle function, brain, skin, cardiovascular, liver, kidney, or even slowing the progression of liver cancer were observed.
The fast track would be to think that by consuming this NAD+, we could increase its levels in cells, but this is not the case since, when ingested, NAD+ is not able to reach cells. However, there are two substances that are capable of achieving this increase of NAD+ at the cellular level. They are nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN), two derivatives of vitamin B3.
In human terms, NMN is used to produce nicotinamide adenine dinucleotide (NAD+). This molecule can be found in all human cells and is vital. Nicotinamide riboside (NR) or niagen is an alternative form of vitamin B3 (niacin) that occurs naturally in trace amounts in milk.
The most notorious difference between NMN and NR is size. NMN is larger, so it needs to be broken down in order to fit the cell. NR is believed by some to be more efficient.
