This week the company selling NMN (nicotinamide mononucleotide) is Purovitalis. This is the Purovitalis home page, it is a Shopify store, it looks good, but then that isn’t difficult now with simplicity of the plug and play templates that Shopify, WooCommerce, Wix etc. offer. Where a poorly designed website may be an indicator of a less than reputable company, a well-designed site just means Purovitalis paid someone on Upwork or Fiver a couple of hundred dollars to put it together, it is not a reflection on the quality of the product. This list of publications here is meant to lead you to believe that Purovitalis have had their product featured, in a positive light, in one of their articles maybe. This is Longevity Technology (https://longevity.technology/), you can see here that there is a top page banner advert, this would be costing Purovitalis a pretty penny I am sure, and if you search Purovitalis on their site, no matches. This is an attempt at social proof, but really, is just misleading marketing. And just below that the statement “Made in Europe” – as we know all know commercial NMN is made in either China or India – so more misleading marketing information from Purovitalis.
Purovitalis have had their site for about 2 and a half years, which is a decent amount of time, long enough to research and build a catalogue of well-established longevity products, let's take a quick look at their shop, so 5 products: NMN, Resveratrol, spermidine, quercetin, and Vitamin D3, but not with Vitamin K2 – TMG would - IMHO - be a no-brainer. Compare Purovitalis to ProHealth, DoNotAge and Renue by Science who have dedicated information and science pages with links to the scientific studies etc. In the marketing copy Purovitalis mention we are the first NMN brand to source Nicotinamide Mononucleotide exclusively from Europe, again this just means that’s where they buy it wholesale, not where it is made; I think they may buy in bulk from someone in Europe and package it in Europe, they can they claim Made in Europe – which is misleading.
Scroll a little lower on the Purovitalis website and there are some product details, mostly okay, just this comment under Fast Dissolving & Absorption – it reads “Our Purovitalis NMN powder is ready to be taken sublingually, which is known to provide the fastest supplement absorption. Taking Purovitalis NMN powder sublingually provides an instant absorption of the supplement into the bloodstream.” There are no studies that show sublingual is superior to oral administration (misleading), and if it is why do they sell liposomal capsules too? It’s all down to convenience and personal preference at the moment.

Discovered by David Sinclair PhD, NMN or Nicotinamide Mononucleotide is the pre-cursor to NAD+ (Nicotinamide Adenine Dinucleotide), NAD is found in nearly all our living cells. NAD is essential for sustaining life. Dr. David Sinclair (a professor at Harvard Medical School) once said if we suddenly didn’t have NAD we would be dead within eight seconds.’ Unfortunately, as we age our NAD levels drastically decline, clinical studies have shown that by age of 50 a typical person could have half the NAD+ they did in their youth and by the age of 80 our NAD+ (Nicotinamide Adenine Dinucleotide) levels drop to around 1% to 10% of those enjoyed in our youth. NAD is biologically unstable, which makes it unsuitable for oral supplementation. Fortunately, it is easy to restore our cellular NAD levels using NMN (Nicotinamide Mononucleotide) or NR (Nicotinamide Riboside), which convert to NAD when it is in the cell. These NAD boosting supplements can be purchased from reputable companies such as Do Not Age, Renue by Science and Pro-health Longevity, however there are many unscrupulous companies selling fake NMN, so buyer beware. Dr. David Sinclair confirms that a fascinating aspect of NAD+ is its dual role in protecting against factors that age us. This includes mitigating chemical stress, inflammation, DNA damage and failing mitochondria; all conditions associated with aging. In other words, while a decline in NAD+ levels may negatively influence lifespan and Healthspan (health-span), so restoring NAD+ is increasingly being viewed as a scientific way to promote longevity and a way to combat the diseases of aging too. A rigorous scientific review of NAD+ reveals that its longevity benefits arise from eight different, but interrelated, functions. In 2013 a scientific study; 2-year old mice were tested for muscle wastage, insulin resistance and inflammation, all are, on humans used as indicators of the aging process, they were then given NMN (Nicotinamide Mononucleotide) for 1 week. In all cases, after only 1 week the 2-year old mice were reflecting the expected findings of 6-month old mice in the areas of muscle wastage, insulin resistance and inflammation. In human terms that would be comparable to a 75-year old having the results you would expect from a 25-year old. In a later methodical study, in 2018, again on mice, Dr. David Sinclair’s team at the Harvard Medical School found that the aging of blood vessels had been reversed.

Back in 2017, 962 million people around the globe were over the age of 60, this already large number is projected to double by the year 2050. Consequently, a considerable amount of attention is now focused on the concept of "active" or "successful" aging, an important component of which seems to be an active social life. A new study suggests that frequent socializing may considerably extend the lifespan of older people.
This study, of more than 28,000 people was published online in the Journal of Epidemiology & Community Health. Socializing nearly every day seems to be the most beneficial for a longer life, but most of the evidence for the health benefits of socializing is based on people in Western countries, with little published data on people who live in Asia, this study went some way to addressing that disparity. To try and plug this knowledge gap, the researchers wanted to explore whether the frequency of socializing might be linked to overall survival in a relatively large group of older people. They participants were drawn from the Chinese Longitudinal Healthy Longevity Survey (CLHLS), an ongoing, prospective nationally representative study of older people living independently, which began in 1998. However, data collection on the “frequency of socializing” only started in 2002. The current study focuses on 5 separate waves of data collection up to 2019, involving a total of 28,563 participants with an average age of 89. The participants were asked how often they engaged in social activities, and were told to record their answers as either:
• Almost every day, at least once a week, at least once a month, occasionally or never.
• Information on influential factors was also collected; this included their sex, level of education, marital status, household income, fruit and vegetable intake, lifestyle and poor health.
Survival was tracked for an average of 5 years or until death.

An estimated 55 million people worldwide live with dementia, a number that's expected to rise as the global population ages. To find treatments that can slow or stop the disease, scientists need to better understand the factors that can cause dementia. There is currently no "cure" for dementia. In fact, because dementia is caused by different diseases it is unlikely that there will ever be a single cure for dementia. Research is currently aimed at finding cures for dementia-causing diseases, such as Alzheimer's disease, Vascular dementia, Lewy body dementia and Fronto-temporal dementia. Researchers at Tufts University recently completed the first study examining levels of vitamin D in brain tissue, specifically in adults who suffered from varying rates of cognitive decline. They found that members of the group who had lived with higher levels of vitamin D in their brains had better cognitive function.
Dr. Sarah Booth PhD, co-senior author of the study and Director of the Jean Mayer USDA Human Nutrition Research Center on Aging said "This research reinforces the importance of studying how food and nutrients create resilience to protect the aging brain against diseases such as Alzheimer's disease and other related dementias."
Vitamin D supports many functions in the body, including immune responses and maintaining healthy teeth and bones. Sources of Vitamin, as well as sunlight, include:
• Cod liver oil
• Salmon
• Swordfish
• Tuna fish
• Sardines, and
• Beef liver
Vegans who do not get adequate Vitamin D from sunlight need to consume Vitamin D fortified products such as bread and juice, with the source coming from algae.
Dr. Kyla Shea PhD, an Associate Professor at the Friedman School of Nutrition Science & Policy at Tufts said "Many studies have implicated dietary or nutritional factors in cognitive performance or function in older adults, including many studies of vitamin D, but all of them are based on either dietary intakes or blood measures of vitamin D. We wanted to know if vitamin D is even present in the brain, and if it is, how those concentrations are linked to cognitive decline."

For decades, doctors have warned people suffering from heart rhythm problems to avoid coffee, this mainly out of concern that a caffeine jolt might prompt an abnormal heartbeat.
But a large study, out of the University of California, San Francisco, found that most people can enjoy their morning Joe free from worry, because caffeine doesn't appear to increase most people's risk of arrhythmia.
Dr. Gregory Marcus, Associate Chief of Cardiology at the University of California, San Francisco, and the study’s co-author said "We see no evidence for this broad-based recommendation to avoid coffee or caffeine, there could be some individuals where caffeine is their trigger, but I think the growing evidence is those cases are actually quite rare."
Coffee is one of the most widely consumed beverages worldwide, but its properties, as a stimulant, have prompted many doctors to warn heart patients against drinking it.
However, the results indicate that every additional cup of coffee a person drinks daily might actually lower their risk of arrhythmia by about 3% on average, this according to the study published JAMA Internal Medicine – and there is a link in the description below to that study.
Dr. Gregory Marcus, University of California, San Francisco noted that "The majority of people, even those with arrhythmias, should be able to enjoy their cup of coffee, and maybe there are some people for whom caffeine or coffee may actually help reduce their risk."
To see whether caffeine really can cause the heart to race or beat abnormally, Marcus and his colleagues analyzed data from more than 386,000 people participating in a long-term British health study.
The researchers said that of that large group (of 386,000 people) only around 17,000 people (so around 4.4%), developed a heart rhythm problem during an average follow-up of 4½ years.
All participants were asked about their coffee consumption when they entered the study.
The researchers then compared their response to their likelihood of developing an abnormal heart rhythm later on down the line.
The result: There was no link at all between caffeine and heart rhythm disturbances, even when researchers took into account genetic factors that might influence the way individuals metabolize caffeine.

Normally, tests to accurately measure VO2 Max, a key measurement of overall fitness and an important predictor of heart disease and mortality risk, require expensive laboratory equipment and are mostly limited to elite athletes. This new method uses machine learning to predict VO2 Max, the capacity of the body to carry out aerobic work, during everyday activity, without the need for contextual information such as GPS measurements. In what is by far the largest study of its kind, the researchers gathered activity data from more than 11,000 participants in the Fenland Study using wearable sensors, with a subset of participants tested again seven years later. The researchers used the data to develop a model to predict VO2max, which was then validated against a third group who carried out a standard lab-based exercise test. The new model showed a high degree of accuracy compared to lab-based tests, and outperformed other approaches. Some smartwatches and fitness monitors currently on the market claim to provide an estimate of VO2max, but since the algorithms powering these predictions aren't published, and are subject to change at any time, it's unclear whether the predictions are accurate, or whether an exercise regime is having any effect on an individual's VO2max over time. The Cambridge-developed model is more robust, transparent and provides accurate predictions based on heartrate and accelerometer data only. Since the model can also detect fitness changes over time, it may also be useful in estimating fitness levels for entire populations and identifying the effects of lifestyle trends on longevity. The measurement of VO2max is considered the 'gold standard' of fitness tests. Professional athletes, for example, test their VO2max by measuring their oxygen consumption while they exercise to the point of exhaustion. There are other ways of measuring fitness in the laboratory, but these require specialized equipment. Additionally, strenuous exercise can be a considerable risk to some individuals.
Dr. Soren Brage, of Cambridge University's MRC Epidemiology Unit, and co-author of the study said "VO2max isn't the only measurement of fitness, but it's an important one for endurance, and is a strong predictor of diabetes, heart disease, and other mortality risks. However, since most VO2max tests are done on people who are reasonably fit, it's hard to get measurements from those who are not as fit and might be at risk of cardiovascular disease."

Contrary to popular belief, not all vegan and vegetarian diets are healthy - and one consequence of a low-quality plant-based diet could well be poorer mental health. New research has found that plant-eaters with a taste for processed foods are more susceptible to depression than their peers who have diets that are high in fresh produce. Megan Lee, a researcher in nutritional psychiatry at Bond University, said the findings were significant given the increasing popularity of vegan and vegetarian lifestyles - and the proliferation of packaged foods that now target those markets.
Megan Lee PhD, a Nutritional Psychiatrist and Senior Teaching Fellow at Bond University said "There is a general perception that following a plant-based diet is inherently healthy, but like any diet it comes down to what you put in your mouth. Many of the products are high in refined vegetable oils and refined grains, salt and sugar. Vegans and vegetarians are not automatically eating heaps of fruit and Veg because there are all these products out there that are fully processed, fully refined. People may inadvertently be consuming high levels of processed plant foods which are a known risk factor for increased depression."
For the study, researchers examined the diet quality and mental health of 219 vegans and vegetarians in Australia. The participants were asked to complete questionnaires based on recognized measures of diet quality and depression. Those with diets high in fresh fruit, vegetables, nuts, seeds, legumes and whole grains were found to be at lower risk of depression compared to fellow vegans and vegetarians with low quality diets. Over 300 million people globally experience the symptoms of depression. Mood disorders, including depression, are significant predictors of suicide and suicidal thoughts, and are responsible for over 800,000 global suicide-related deaths per year. In Australia, 1 in 10 people (that’s 10.4% of the population) report experiencing symptoms of depression. In younger Australian adults, the prevalence rate of depression is higher at 13.9%; with suicide listed as the leading cause of death for individuals aged between 15 and 44.

Renue by Science received applications from approximately 120 people whose average deep sleep (in minutes) was calculated using their own tracking devices.
Renue by Science only selected people who provided:
o Firstly, enough data to determine an accurate baseline average
o And secondly, people who were deemed to be low on deep sleep (that was assessed to be between 0 and 45 minutes per night).
Renue by Science instructed the participants to follow the following protocol:
• Day 1: Take 1 x Liposomal Apigenin capsule 1-4 hours before bed (each Lipo Apigenin capsule contained 35mg of Apigenin).
• After 3 days, so on day 4 increase to 2 capsules, still taken 1-4 hours before bed, that was then a dose of 70mg per night
• If there were no improvements seen on their deep sleep monitor, they were asked to increase the dose to 3 x Liposomal Apigenin capsules, again 1-4 hours before bed, which was then a dose of 105mg per night).
There were added requirements that the participants were also told to follow:
• Refrain from increasing their normal intake of alcohol or caffeine.
• And also to try not to vary exercise levels too much as too much exercise can increase the Resting Heart Rate, and decrease Deep Sleep and Heart Rate Variability.

"Zombie cells" that contribute to age-related diseases also help heal damaged tissues, so wiping them out could come with major downsides. The zombie cells, scientifically known as "senescent" cells, are cells that stop multiplying due to damage or stress but don't die; this according to the National Institute on Aging. These zombie cells then release a slew of molecules that summon immune cells and this in turn spark inflammation.
The immune system regularly clears these zombies from the body, but with age, the system becomes less efficient; and as a result, the dysfunctional cells accumulate. This accumulation produces more inflammation, which in turn contributes to the development of age-related diseases such as cancer, Alzheimer's disease and osteoarthritis.
But zombie cells aren't entirely bad. This new study, conducted in lab mice and on human cells, suggested that senescent cells helped repair lung tissue after damage by encouraging stem cells to grow. Killing these zombie cells with Dasatinib, a cancer treatment, and Quercetin, a naturally occurring polyphenol – a drug duo that's been studied as a potential treatment to combat aging and age-related diseases - was found to somewhat disrupted this repair.
Tien Peng MD, an associate professor of pulmonary, critical care, allergy and sleep medicine at the University of California, San Francisco said
"We are not the first lab to implicate senescence as a wound-healing process. A 2014 study in the journal Developmental Cell found that zombie cells helped mend wounds in the skin and that their repair can also be disrupted by senolytics. This suggests that using senolytics could come at a cost, so the drugs will have to be designed to block zombie cells' bad effects without disrupting their good ones.”
To find senescent cells in the lung, researchers genetically modified mice to carry a glowing protein on the gene that codes for the protein "P16," which is overactive in many senescent cells. Whenever a cell switched on the gene, it would also churn out fluorescent proteins and start to glow. The researchers used this technique to "really amplify this signal," Professor Peng said, and thus revealed cells that carry low levels of p16; cells that may have otherwise escaped notice. Glowing cells appeared in mice's lungs shortly after birth, and their numbers increased over the rodents' life spans. The cells included fibro-blasts, which make connective tissue, as well as immune cells; they reside within a sheet-like tissue called the "basement membrane." The basement membrane is a sort of gatekeeper, it singles out and blocks harmful chemicals and pathogens from entering the lungs. But, at the same time it also allows oxygen to pass unhindered into the bloodstream.
