Discovered by David Sinclair PhD, NMN or Nicotinamide Mononucleotide is the pre-cursor to NAD+ (Nicotinamide Adenine Dinucleotide), NAD is found in nearly all our living cells. NAD is essential for sustaining life. Dr. David Sinclair (a professor at Harvard Medical School) once said if we suddenly didn’t have NAD we would be dead within eight seconds.’ Unfortunately, as we age our NAD levels drastically decline, clinical studies have shown that by age of 50 a typical person could have half the NAD+ they did in their youth and by the age of 80 our NAD+ (Nicotinamide Adenine Dinucleotide) levels drop to around 1% to 10% of those enjoyed in our youth. NAD is biologically unstable, which makes it unsuitable for oral supplementation. In a later methodical study, in 2018, again on mice, Dr. David Sinclair’s team at the Harvard Medical School found that the aging of blood vessels had been reversed. The science showed that mice that were given NMN (Nicotinamide Mononucleotide) had restored blood vessel growth and had increased muscle endurance by up to 80%. Dementia remains largely undetected, even in high-income countries such as Canada where rates of undetected cases exceed that of 60%. But why?
Belief that cognitive decline is normal in elderly people, and the lack of knowledge of dementia symptoms, and of diagnostic criteria among medical doctors have been identified as the main culprits of missed cases and delayed diagnosis. Age-related memory losses should not be shaken off as just part of normal aging. Occasionally forgetting where we parked the car or where we left our keys can happen to anyone, but when these situations become frequent it's important to seek medical advice. for many, these lapses in memory will not result in the development of dementia, but in others, these declines constitute an early warning sign. Research has shown that people with mild changes in cognition are at a greater risk of developing dementia later in life. It has been demonstrated that the disease process starts decades before the appearance of symptoms such as memory loss. The scientific community recognizes that interventions which aim to slow down or prevent disease development are more likely to be effective when initiated early in the disease course. Despite this, protocols for early detection are not standard in the medical community, in part because significant gaps remain in our understanding of dementia.
In her research, Stefanie Tremblay used advanced brain MRI methods to characterize brain health in older adults who are at high-risk of developing dementia. Her goal is to identify new biomarkers of early pathology, which could lead to improved detection methods in the future.
She stated that “The proportion of senior Canadians is growing in our population. Dementia is strongly associated with aging, so the number of Canadians diagnosed with dementia, including Alzheimer's, is expected to rise considerably in the next few decades, reaching an expected 1.7 million Canadians by 2050.”

According to the CDC, between 6.7% to 36.9% of Americans take vitamin D supplements, that’s depending on their age group, I’m guessing this must be similar in most 1st world countries. This makes it the second most popular dietary supplement in the USA. Vitamin D is essential for calcium absorption in the gut, it also regulates calcium, phosphorus, and glucose metabolism, and it supports bone growth. Many of our genes are controlled by Vitamin D, and the vitamin D receptor is present in many of the body’s tissues. It is not surprising, then, that low levels of Vitamin D have been associated with multiple disorders, including cardiovascular problems, diabetes, cancer, and numerous inflammatory disorders.
Due to Vitamin D’s essential roles in the body, much research has been done on its impact on mortality risk. However, the reported results are inconsistent. This inconsistency, the limitations of previous research, and the emergence of new studies has prompted researchers to analyze existing, and new research, into Vitamin D supplementation. IN this new study researchers looked at the results of randomized controlled trials that were published between 1983 and 2022. They wanted to know if taking vitamin D supplements affected overall mortality and wanted to know if it was linked to deaths from cardiovascular disease. So, the studies that were included in this analysis single out all-cause mortality, cardiovascular mortality, non-cardiovascular mortality, and cardiovascular morbidities: the last one on the list are cardiovascular issues that do not necessarily lead to death.
Studies included in the analysis investigated vitamin D supplementation both, with and without, calcium added. The total number of all participants in the analyzed studies was more than 160,000, with a mean age of 66. However, it didn’t include pregnant or lactating women, frail or elderly patients, or those with serious medical conditions, such as stroke, COVID-19, and HIV. The studies in this analysis varied in their design and in their quality.

As you may know, if you have watched my previous step count presentations, you will know that 10,000 steps a day is not a scientifically derived number, in all cases, studies have shown, beneficial numbers are less than 10,000. And now, the number of steps you should take every day, to start seeing benefits to your health, is also lower than previously thought; this is according to the largest analysis conducted to date.
The study, published in the European Journal of Preventive Cardiology found that walking at least 3,967 steps a day started to reduce the risk of dying from any cause, and Only 2,337 steps a day reduced the risk of dying from cardiovascular disease. This new analysis of 226,889 people from 17 different studies around the world has shown that the more you walk, the greater the health benefits, however An increase of 1000 steps a day was associated with a 15% reduction in the risk of dying from any cause, and an increase of just 500 steps a day was associated with a 7% reduction in dying from cardiovascular disease. In line with the previous studies I have covered, the researchers also found that if you were to walk more than the lower numbers previously shown, the health benefits continued to increase.
Professor Maciej Banach from the Department of Preventive Cardiology and Lipid-ology at the Medical University of Łódź, Poland said, "Our study confirms that the more you walk, the better. We found that this applied to both men and women, irrespective of age, and irrespective of whether you live in a temperate, sub-tropical or sub-polar region of the world, or a region with a mixture of climates. In addition, our analysis indicates that as little as 4,000 steps a day are needed to significantly reduce deaths from any cause, and even fewer to reduce deaths from cardiovascular disease."
There is obviously strong evidence that a sedentary lifestyle contributes to an increase in cardiovascular disease and a shorter life. Studies have shown that insufficient physical activity affects more than a quarter of the world's population. More women than men (32% versus 23%), and people in higher income countries when compared to lower income countries (37% versus 16%) do not complete enough physical activity throughout the average day.

It appears to be VERY good news for those of you who want to keep fit, and live longer, but who don't like playing sport or going to the gym. This new research shows that just 3 to 4 one-minute bursts of huffing and puffing during daily tasks is associated with large reductions in the risk of premature death, particularly death from cardiovascular disease.
The study, that was Published in Nature Medicine, was led by the University of Sydney's Charles Perkins Center. In a first of its kind, the study accurately measured the health benefits of what researchers have termed "Vigorous Intermittent Lifestyle Physical Activity" or VILPA for short. VILPA is defined as very short bouts of vigorous activity (between 1 or 2 minutes). Things we do with gusto each day. These could be activities like running for the bus, bursts of power walking while doing errands or playing high-energy games with the kids. The researchers found that just 3 to 4 one-minute bouts of VILPA every day is associated with:
• Up to a 40% reduction in all-cause and cancer-related mortality, and.
• Up to a 49% reduction in death related to cardiovascular disease.
Emmanuel Stamatakis, lead author of the study and Professor of Physical Activity, Lifestyle & Population Health at the University of Sydney's Charles Perkins Center said, "Our study shows similar benefits to high-intensity interval training (HIIT) can be achieved through increasing the intensity of incidental activities done as part of daily living, and the more the better.
A few very short bouts totaling three to four minutes a day could go a long way, and there are many daily activities that can be tweaked to raise your heart rate for a minute or so."
The vast majority of adults, aged 40 and over, do not take part in regular exercise or sport. But Professor Stamatakis said, the study reveals how incidental physical activity can overcome many of these barriers. He noted that, "Upping the intensity of daily activities requires no time commitment, no preparation, no club memberships, no special skills. It simply involves stepping up the pace while walking or doing the housework with a bit more energy.”
The team of researchers used wrist-worn tracker data from the UK Biobank Study.

Living to be older than 100 is indeed a rare feat for any human being. This study has shown that, in part, centenarians could have their gut microbiome to thank. The study, of Japanese people, who have lived over a century found they harbor distinct groups of bacteria in their gut, which generate unique compounds and might even ward off infections and other environmental stressors.
Centenarians, generally speaking, are less inclined to suffer from age-related diseases and are somehow capable of thwarting infectious diseases too. This study now seems to show that the gut microbiome – the billions (with a B) of micro-organisms living in our digestive tract – could help to explain the reason why. Many people still think that the secret to a long life is written in our genes, and only in our genes, with stories of relatives who smoked heavily from the age of 14 and lived to be 101.
But genetics accounts for less than 30% of our longevity, this then leaves the door wide open to many other factors that, some of which we can control and that change over time, such as:
A healthy diet
High quality sleep
Regular quality exercise
Meaningful social interaction (not scrolling through social media), and now also, our billions of gut bacteria.
The researchers recruited 160 centenarians from across Japan who had racked up an impressive average age of 107 years.
They then compared the bacterial communities found in their fecal samples - to the bacteria of another 112 elderly people in their late 80’s, and also to those of 47 younger people.
The researchers, led by micro-biologist Yuko Sato from Keio University School of Medicine in Tokyo, were looking for differences in their gut microbiomes.
That is to say, the types of bacterial species present, and the sorts of compounds they produced.
Previous studies of centenarians, conducted in Sardinia, in Italy found that people aged over 100, when compared to younger people, had a higher diversity of core micro-biota species living in their intestines.

It may shoot through your hands while typing or flare up in your knees when getting out of the car or out of the bath, but wherever the pain is, over 32 million Americans, and an estimated 10% of all men and 18% of women over the age of 60, worldwide, are living with osteoarthritis, and the pain and physical limitations that it brings with it. To reduce that constant pain, patients living with the crippling condition are often told by their doctors to simply exercise. It sounds simple, but people with osteoarthritis may experience pain when they start to move more, which can be a deterrent to taking up, or sticking with, any exercise program.
Daniel Whibley PhD, an Assistant Professor of Physical Medicine & Rehabilitation at the University of Michigan said "Pain during movement is an important reason why this population isn't more active, and we need to identify ways we can help to change this.
Otherwise, they may end up in a loop of pain and inactivity that we know can lead to disability later down the line."
A growing body of research suggests that maintaining good sleep health, more specifically high-quality sleep for an appropriate duration, in patients with osteoarthritis may reduce their pain and discomfort.
There is also strong evidence supporting links between sleep and being physically active.
Professor Whibley stated that "If you're sleeping well, you're more likely to be able to move more the next day or stick with a planned exercise program. And those who are physically active during the day are more likely to get a good night's sleep.”
Professor Whibley's team looked to develop a new intervention that brought together insights from previous research that supports these relationships.
What hasn't been investigated yet is whether simultaneously targeting both sleep and physical activity - and the optimal balance between them – will result in better pain outcomes. With his team of researchers, Professor Whibley introduced the concept of a hybrid sleep-exercise program, that observed focus groups composed of people living with BOTH OA related pain and sleep disturbances.
The sleep component of this intervention is an automated program called Sleepio. Sleepio delivers cognitive-behavior therapy for insomnia over the internet, as well as components focused on sleep education and behavior modifications to sleep routines.

Coffee is one of the world’s most widely consumed beverages, and numerous studies have shown that having higher coffee consumption is linked to decreased rates of mortality, as well as decreased rates of neurological and metabolic diseases, including Parkinson's, Cardiovascular disease, and Type 2 diabetes. This new study adds to that body of evidence, by saying that having a second cup may actually be extremely beneficial for regular coffee drinkers, let’s find out why.
Stephen Safe, PhD – Professor of Toxicology at Texas A&M University, and co-author of the paper said, "There is evidence that higher coffee consumption is associated with lower rates of colon and rectal cancer, as well as breast, endometrial and other cancers, although there are conflicting reports on its benefit for some of these cancers.”

The research was conducted in the Safe Lab and the Chapkin Lab, at the Texas A&M University.
Two of the primary contributors to the study were Professor Robert Chapkin and Laurie Davidson PhD, she is a Department of Nutrition researcher who works in the Chapkin Lab.
Professor Robert Chapkin, Head of the Chapkin Lab at Texas A&M University said, "The mechanisms associated with the chemo-preventive or chemo-therapeutic effects of more than 1,000 individual compounds in roasted coffee are complex and may vary with different diseases."
Some of these mechanisms may be related to pathways that target oxidative stress or pathways that induce Reactive Oxygen Species (ROS) to kill diseased cells. There is also evidence for the involvement of receptors in addition to contributions from epigenetic pathways and the gut microbiome.

There is no debate that getting good quality sleep at night plays a substantial role in supporting your heart, your general health, and your overall wellbeing. And now, new research has shown it could also affect how long you live. This latest study found that young people who have higher quality sleep habits are incrementally less likely to die early. Moreover, the data suggest that about 8% of deaths from all causes could be attributed to poor sleep patterns.
Frank Qian (chein), MD, an internal medicine physician at Beth Israel Deaconess Medical Center and a Clinical Fellow in medicine at Harvard Medical School, and co-author of the study said "We saw a clear dose-response relationship, so the more beneficial factors someone has in terms of having higher quality of sleep, they also have a stepwise lowering of all cause and cardiovascular mortality. I think these findings emphasize that just getting enough hours of sleep isn't sufficient. You really have to have restful sleep and not have much trouble falling and staying asleep.”
For their analysis, the team included data from 172,321 people who participated in the National Health Interview Survey (the NHIS) between 2013 and 2018. The cohort had average age of 50; with 54% of them being women. About two-thirds of study participants self-reported as being White, 14.5% Hispanic, 12.6% Black and 5.5% as Asian. This National Health Interview Survey is fielded each and every year by the Centers for Disease Control and Prevention (the CDC) and the National Center for Health Statistics. Its aim is to help gauge the health of the U.S. population and includes questions about sleep and sleep habits.
Because researchers were able to link the study participants to the National Death Index, they could examine the association between individual and combined sleep factors, and all-cause - and all cause-specific mortality.
Participants were followed for a median of 4.3 years during which time 8,681 individuals died.
Of these deaths,
• 2,610 (that’s 30%) were from cardiovascular disease.
• 2,052 (that’s 24%) were from cancer, and
• 4,019 (that’s 46%) were due to other causes.
Researchers rated five different factors of sleep quality using a low-risk sleep score that they created based on answers collected as part of the survey.
The Factors
• Achieving the ideal sleep duration of seven to eight hours a night.
• Having difficulty falling asleep, but no more than twice a week.
• Having trouble staying asleep, but no more than twice a week.
• Not needing to use any type of sleep medication; and
• feeling well rested after waking up, but at least five times a week.

A team led by famed, some may say infamous, Harvard Medical School anti-aging researcher Dr. David Sinclair have just published a paper, that shows that a specific mix of chemicals are capable of reversing cellular aging. If it sounds too good to be true, it could well be. So far, the evidence is only in human cells in the lab, it appears that we are a long way from testing this on animal models, and obviously further away from human trials.
Dr. David Sinclair PhD, a professor in the Department of Genetics at Harvard Medical School wrote in the paper "We identify six chemical cocktails, which, in less than a week and without compromising cellular identity, restore a youthful genome-wide transcript profile and reverse transcriptomic age. Thus, rejuvenation by age reversal can be achieved, not only by genetic, but also chemical means."
If this statement is taken out of a cell-level context, which some individuals appear to have done for some reason, this amounts to a very bold claim, given how early in the process this research is. So, it's unsurprising that there's already some controversy surrounding this paper.
The study looked at the way our cells lose information as they age, specifically their epigenetic information. As they get older, our cells go from having unlimited potential when we're embryos, to separating into just one specific cell type, and eventually becoming senescent, running out of functionality, and dying.
In 2012, Shinya Yamanaka and Sir John Gurdon won a Nobel Prize for showing this process can actually be reversed. They found that turning on the so-called Yamanaka factors (OCT4, SOX2, and KLF4) in mammals can turn adult cells back into induced pluri-potent stem cells with unlimited potential. A few years later, a team of researchers showed that even temporarily turning on these Yamanaka factors in mice made the rodents healthier.
Since then, the race has been on to apply the same idea in humans, but without turning cells cancerous, which is a real risk when you give cells the ability to divide endlessly. Doctor Sinclair's lab has already achieved promising early results, they have been able to use Yamanka factors to restore vision in mice and monkeys who had damaged optic nerves.

Researchers have discovered not only that animals age more quickly when they don't have enough of the amino acid taurine in their body. And that taurine supplements can delay aging and increase a healthy lifespan. An international team of researchers found that taurine supplements delayed aging in worms, mice, and monkeys, and increased the healthy lifespan of middle-aged mice by up to 12 percent.
Vijay Yadav, an Assistant Professor of Genetics and Development at Columbia University’s Vagelos (Vaj-il-us) College of Physicians & Surgeons, and senior author of the study said "For the last 25 years, researchers have been trying to find factors that not only let us live longer, but also increase Healthspan, the time we remain healthy in our old age.
This study implies that taurine could be an elixir of life within us, that helps us live longer and healthier lives."
Taurine is found naturally in meat, fish, and dairy products, but not very often in plants.
Humans can synthesize taurine, although nutritional sources are often necessary during early life because newborn bodies are not skilled at producing it. Research on taurine has shown benefits in diabetes and as an antioxidant, but its role in many cases is still unclear.
Professor Yadav stated "We realized that if taurine is regulating all these processes that decline with age, maybe taurine levels in the bloodstream affect overall health and lifespan. We started to ask if taurine deficiency is a driver of the aging process, and we set up a large experiment with mice.”
