Caco-2 Permeability Assay

Caco-2 permeability assay is one of the recommended in vitro methods in the United States FDA guidelines to evaluate drug permeability. It has become the gold standard method to evaluate both the passive and active transport and the absorption of orally administered drugs. The method can be used to assess the bidirectional permeability of a test compound (from the apical to the basolateral side or vice versa). By measuring the transport rate of a compound across a Caco-2 cell monolayer membrane, the in vivo absorption of the compound across the gut wall can be well predicted.
