The film closely follows the plot of Arthur Conan Doyle's classic short story. Sherlock Holmes (Raymond Massey) and Dr. Watson (Athole Stewart) are approached by the distraught Helen Stoner (Angela Baddeley). She lives with her stepfather, the formidable and ill-tempered Dr. Grimesby Roylott (Lyn Harding), at the dilapidated Stoke Moran estate.
Helen is terrified because her sister, Julia, died mysteriously two years prior, just before her wedding. Julia's last words were "The speckled band!" Now, Helen herself is engaged to be married, and strange things have begun to happen at the estate, echoing the events that preceded her sister's death. She hears a low whistling sound at night and has observed odd behaviors from Dr. Roylott, including his strange pets (a baboon and a cheetah) and his insistence that she sleep in the same room where her sister died, which has a bell cord that goes nowhere and a ventilation shaft leading to his room.
Holmes, intrigued by the bizarre circumstances and sensing foul play, takes on the case. He and Watson travel to Stoke Moran to investigate. Through keen observation and deduction, Holmes uncovers Dr. Roylott's sinister plot involving a deadly snake, the "speckled band," which is trained to enter Helen's room through the ventilation shaft and deliver a fatal bite. Holmes must race against time to expose Roylott's scheme and save Helen from the same fate as her sister.
