Simply adrenaline drops. Now these drops can add pigment in normal eyes. But Magnussen and Mengele were not studying normal eyes, they were studying a defect, a genetic syndrome which didn't have a name or identification yet. However, the pigment did change in the macula. That is the brown pigmented spot you can see in eye exam photos.
