95. Amlexanox - A Repurposed Anti-Inflammatory

308

Amlexanox (2-amino-7-isopropyl-5-oxo-5H-chromeno[2,3-b]pyridine-3-carboxylic acid), an inhibitor of inflammatory mediators and a phosphodiesterase inhibitor, has obvious therapeutic potential. Amlexanox also inhibits cell proliferation and migration. It inhibits PI3K/Akt/MAPK and NF-κB signalling. This could bring about its phenotypic outcome on cell proliferation and migration in vitro. The cell cycle is inhibited in G1 via the operation of the IκB kinase (IKK)-related kinases TBK1/IKK-epsilon, possibly also the Akt/MAPK pathway (Moser et al., 2016). Given that this pathway can inhibit MMPs, the inhibitory effects of amlexanox on cell migration have become more revealing of the involved mechanisms. That it can promote the depolymerisation of actin filaments is supplementary evidence for its effect on cell motility.

Chat:
20:07:45 From Paula Dow To Dr. Tom Lewis(privately) : I’m taking about 1/2 t of MB per day with 1/2 t buffered vit C. She it be taking more than once/day?
20:09:08 From Paula Dow To Dr. Tom Lewis(privately) : She I be careful about doing anything that makes my heart beat faster? Like working out? Although I really don’t have the energy to do that for the last few weeks. HB is still between 80-85
20:10:34 From Steve : Would Amlexonax users lose weight with the inhibition of mTOR?
20:10:58 From Paula Dow To Dr. Tom Lewis(privately) : is hemolysis an concern/issue for me?
20:12:12 From Steve : Replying to "I have been supporti..."

Excellent Stefan. We all need parallel systems to replace (eventually) the relics of the past.
20:13:12 From Doris Fellenz : Martha, is a lot of what he says going over your head or is it me. He is talking to fast. And that is a drug. What are we discussing a drug for of chemicals?
20:13:15 From Steve : Does it balance Thi/Th2?
20:13:46 From Doris Fellenz : full of chemicals or am I mistaken?
20:14:05 From Maura : God does not create bad genes do you think

What if these are protective genes
20:14:09 From Steve : Replying to "full of chemicals or..."

Which link are you looking at Doris?
20:14:43 From Dee : Would Amlexanox benefit a person who has developed autoimmune arthritis especially scleroderma?
20:15:46 From Maura : Genes do not create pandemic
What else is going on
20:15:56 From Steve : No, that IS a bad thing
20:16:25 From Winthrop Harewood : Would it be also or can be also be considered gene therapy?
20:16:32 From Martha Stark, MD : Apparently, amlexanox was discontinued in the US in 2017 because of “adverse side effects.” What a shame! What are they thinking the adverse side effects were?
20:17:10 From Steve : I would use curcumin and boswellia for cox/lox
20:19:48 From Doris Fellenz To Dr. Tom Lewis(privately) : I just read the following: Is amlexanox available in USA?
This drug has been discontinued in the U.S.
20:20:23 From iPhone : Has it shown any benefit for Rheumatoid Arthritis?
20:20:38 From Maura : Infection by injection
20:20:42 From Steve : If I'm not mistaken, the th1 / th2 theory is just that.
20:23:15 From Maura : What is Root Cause ?
20:23:32 From Dr. Tom Lewis : “We found that minocycline interacted with CD4+ T cells and accelerated CD4+ T cells to differentiate into Th2 and Treg cells, while suppressing the their differentiation into Th1 and Th17 cells and reducing neuroinflammation and white matter injury after ICH”
20:23:59 From Mike Boehmer : If anyone has further questions about the mechanisms and use of amlexanox or the other drugs mentioned please contact me at mboehmer@amexpharmacy.com. Dr Halasa and I are partners in CDPMCs and we can reply together for you.
20:25:19 From Stefan Hartmann, PA-C : I just sent an Rx for amlexenox to you Mike for a landscaper guy who gets SEVERE reactions to grass. It knocks him on his ass for days.

Mike, will negative effects of this drug be discussed tonight?

Sorry but they are disaster

and always have been.
20:26:21 From Dr. Tom Lewis : Our results argue for a reduction of both systemic and local Th17 response after prophylactic, low-dose clarithromycin administration in patients with non-CF bronchiectasis, suggestive of a potential anti-inflammatory and/or immunomodulatory action.
20:27:16 From Dr. Tom Lewis : I am showing these because amlexanox is not the only treatment for inflammatory disease - but it may be complementary or potentially more powerful? Or have fewer side effects.
20:27:23 From Stefan Hartmann, PA-C : Replying to "Vaccines

The corrupt the genetics that dr. Halasa is talking about. Amlexenox can help these people who have activated immune system from these epigenetic mutations that happens from the poisons patients are exposed to

Low dose antibiotic chronically is not something I would want for myself.

Interesting! Macrolides do have immune effects as you mention, tacrolimus is a macrolide.
20:29:26 From Maura : What about plasmids in the “vaccines”
20:30:00 From Dr. Tom Lewis : Stefan: Often at low dose, even though it is called an antibiotic, it does not express that activity. I can point you to the world expert on this topic - Mark Nelson - I have his lecture on my channel. He has investigated repurposed low dose ABx as anti-inflammatories and found at the anti-inflammatory level they do not express antibiotic activities…. Dose make the poison or cure.
20:30:22 From Mike Boehmer : Replying to "If anyone has furthe…"
We can. There really aren’t many that we have seen so far
20:30:23 From Stefan Hartmann, PA-C : Ahh I see
20:30:50 From Stefan Hartmann, PA-C : Replying to "If anyone has furthe..."

I have not seen any side effect with amlexenox so far. Have given it to about 10 patients so far
20:31:16 From iPhone : Reacted to "Stefan: Often at low…" with ❤️
20:31:25 From Dr. Tom Lewis : Minocycline, for example, is neuroprotective and should be standard of care post stroke…
20:31:28 From Dr. Tom Lewis : The favorable effects of minocycline in a therapeutic window of 72 hours can be due to the various minocycline mechanisms of action during stroke. It can be hypothesized that the anti-apoptotic effect, which has been shown in minocycline use,5,6 will appear in a later poststroke stage after more than 48 hours.
20:31:39 From Steve : Replying to "If anyone has furthe..."

Are there blood/etc. tests that would determine who might benefit from this drug?
20:31:59 From Stefan Hartmann, PA-C : Replying to "If anyone has furthe..."

I use the CBC and look at EOS
20:32:08 From Dr. Tom Lewis : Just an example of an Abx that has important other effects.
20:32:09 From Maura : Turtles all the way down

Yes, Cyrex labs lymphocyte mapping and inflammatory cytokine panel can reveal baseline immune polarization and changes would be one example
20:34:05 From Dr. Tom Lewis : I know Dr. Hallasa speaks at a high level but hang in there. So did my mentor - but you get more from this type of information long-term.
20:34:05 From Mike Boehmer : Replying to "If anyone has furthe…"
Also, the big thing is mast cell activation. He hasnt talked about that
20:34:30 From Dee : Who can prescribe and monitor Amlexanox?
20:34:35 From Steve : Replying to "I know Dr. Hallasa s..."

IMO, this is just right level.
20:34:45 From Joy Gentle : @Stefan Hartmann, PA-C Will you chat more about EOS as indicator for Amlexanox?
20:34:49 From Stefan Hartmann, PA-C : I’m in another zoom right now with Dr. Saleeby he says it amlexenox can work for MCAS as well
20:35:23 From Mike Boehmer : Yes MAS is the biggest use other than AMPK
20:35:54 From Dr. Tom Lewis : Dee - see the string. I think Mike at mboehmer@amexpharmacy.com. can help - either prescribe or direct to a source.
20:38:04 From Steve : No way to excape est. dominance is these days with 85,000 toxic chemicals in our soup. Using herbals such as Chrysin to maintain healthy estro levels is a possibility.
20:38:32 From Stefan Hartmann, PA-C : Replying to "No way to excape est..."

Nonsense. Drink good water. Take iodine
20:38:57 From Steve : Replying to "No way to excape est..."

Too reductionist.
20:39:28 From Martha Stark, MD : Reacted to "Too reductionist." with 👍
20:40:20 From Dr. Robert Quinn : Replying to "No way to excape est…"
Steve! I agree 100%! That’s why virtually everyone needs to detox!

Yes, humic/fluvic is one of many to use.

PectaSol another
20:40:58 From Dr. Tom Lewis : I am interested in this material because reducing inflammation is a challenge - especially in the brain. However, it is always important to always ask this question - That I learned from Dr. Thomas Sowell - “Compared to What?”
20:41:26 From Dr. Tom Lewis : Bentonite clay and high dose vitamin c pulses is what I do - along with the fulvic/humic liquid

For brain inflammation, have you considered oxytocin nasal spray, methylene blue, ozone or HBOT? We have had lectures in support of those as well. Melatonin too.
20:43:45 From Dr. Robert Quinn : Replying to "I just sent an Rx fo…"
I’m sure you’ve tried high dose Bromelain and Quercetin, right?

Yes, two excellent choices.
20:44:35 From Dr. Tom Lewis : I have not tried the oxytocin nasal spray for that. I will investigate - I have dozens who need something more…
20:46:40 From Maura : Is there any bias here
20:47:21 From Stefan Hartmann, PA-C : Tom I really am interested in your number crunching program.

20:52:38 From Tina Bruno : Replying to "Is there any bias he..."

Pharma
20:54:41 From Dr. Tom Lewis : Steve.. What I am saying is we have about one thing and its benefits - but seldom is it compared to something with a similar mechanism that has been around for a while. The drug companies are notorious to comparing their efficacy to nothing or something bad. We always need to think about other options. This drug is not the only one that does what Dr. Halasa is talking about - not to say it is bad or inappropriate….. That’s why I sent out some links to the drugs that Dr. Trempe used.
20:55:46 From Steve : Replying to "Is there any bias he..."

I believe he's connected to compounding Pharma
20:55:54 From Dr. Tom Lewis : I do not think there is bias - he is presenting on amlexanox - useful - However, we need to be aware of options and comparisons - but those types of studies cost $$$$$$$$$$$$
20:56:04 From Doris Fellenz : Did they band Amlexanox in 2017 because it cures cancer?
20:56:40 From Steve : MB patients need to have the chemical activated.
20:57:44 From Steve : Do you suggest the MB customers activate the MB?
20:58:20 From Steve : Replying to "Did they band Amlexa..."

Does it really 'cure' cancer?
20:58:21 From Dr. Tom Lewis : Not banned - just “discontinued”
20:58:43 From Dr. Tom Lewis : Methylene blue absorbs red light so intensely, it is probably activated in the present of light.

yes, 660nm
20:59:13 From Mike Boehmer : We typically don’t but you could reduce it with Vit C, but it will re-oxidize in the blood most likely. The dose is too low to have it make a huge effect

I used to do laser IV after the MB IV

Are there reputable studies that show the efficacy of low dose MB? All the practitioners/researchers I know use higher doses
21:01:31 From Steve : Replying to "I sit under my halog..."

Have you checked out the wavelength spectrum of halogen?
21:01:57 From Steve : Replying to "I sit under my halog..."

I use a red.light watch with 660nm LEDs
21:02:06 From Dr. Tom Lewis : Steve: very intense output at 660/670 as does sunlight
21:02:35 From Steve : Replying to "Steve: very intense ..."

Good to know. Haven't looked at the spectrum myself.

21:03:08 From Mike Boehmer : A couple of points, Dr Lewisnis correct, Amlexanox was not removed from the market, it simply did not sell well for apthous ulcer and was dc by the manufacturer. This was the 80s/90s and they did not fully understand it
21:03:15 From Dr. Tom Lewis : That shows the spectrum - full spectrum similar to sunlight

Excellent, tnx!!
21:03:33 From Linda Anderson : Look into the MB chemistry. Some use a non-blue version, it is diff chemically and is not blue. Dr Levy may use that. it's similar, not the same. I use 20 drops/ 10mg in water.... blue tongue. just started. it is helping urinary issues
