1. How mRNA Vaccines and Vaccine Breakthrough Infections Drive Viral Immune Escape Dynamics

🔹 The development of herd immunity to COVID-19 has been hindered by mRNA vaccines and vaccine breakthrough infections of new Omicron variants. Instead, highly vaccinated populations are exerting herd immune pressure on viral infectiousness.

🔷 During a pandemic of an acute self-limiting viral infection, the best way to protect ourselves is not through vaccination but through training of our innate immune system. Training our body's innate immunity is the best way to guard against potentially highly virulent SARS-CoV-2 variants. Although both cell-based innate immunity and virulence-inhibiting, non-neutralizing antibodies effectively protect against (severe) disease caused by any SARS-CoV-2 variant, only trained cell-based innate immunity has a durable sterilizing effect. This is why healthy unvaccinated individuals are now largely protected from symptomatic infection and critically contribute to herd immunity.

3. Is Covid Vaccine Protection Durable?

🔷 People who have been vaccinated against COVID-19 are still benefiting from protection against severe illness, but relying solely on weak and short-lived antibodies means that protection is suboptimal and only temporary. Re-exposure to the virus or a vaccine booster may initially sustain protection, but not without increasing immune selection pressure on viral pathogenicity.

As the latter are weak and not long-lived, they exert immune selection pressure on the very pathogenicity of the virus, which could present us with an even greater health threat in the future. 🔷 Are our antibodies still effective against new viruses? Scientists are working hard to understand how our bodies respond to new virus variants, like the Omicron XBB, to ensure we remain protected.

6. Omicron Causes Enhanced Large-Scale Immune Escape Dynamics

🔷The advent of Omicron caused viral immune escape dynamics to reach a point of no return, transforming the immune escape pandemic into an 'inescapable' self-catalyzing chain of enhanced immune escape, instead of driving it into endemicity.

🔷 mRNA-based Covid-19 vaccines are considered problematic from both an individual and public health perspective due to their ability to shift the focus of the immune response to poorly immunogenic domains of spike protein.
However, individuals who have received a single dose of an mRNA-based COVID-19 vaccine or a maximum of two doses of a non-mRNA vaccine, and subsequently experience a symptomatic breakthrough infection, are believed to retain the ability to train the broadly protective capacity of their cell-based innate immune system.
