Urolithin A is a microbial metabolite of ellagic acid, produced in the gut following consumption of ellagitannins. Ellagitannins are bioactive polyphenols in fruits and nuts such as pomegranates, raspberries, strawberries, and walnuts. These polyphenols are poorly absorbed in humans, but their metabolization by the gut microflora yields a series of urolithins. Urolithin A has been shown to have neuroprotective effects against Alzheimer's disease, through its anti inflammatory effects. Recently, neuro inflammation driven by the brain's microglia has been shown to play an essential part in the pathogenesis of Alzheimer's disease. Alzheimer's disease is a neurodegenerative disease characterized by the accumulation of amyloid beta that activates microglia. Activated microglia has been reported to produce pro inflammatory factors such as interleukin 1 beta and interleukin 6 in the brain. Both of these proteins increases amyloid precursor protein expression and amyloid beta deposition in animal models of Alzheimer's disease. It has been reported that urolithin A, which exhibits anti inflammatory properties, may reduce the risk for Alzheimer's disease by more than 50 percent. However, the underlying mechanisms are still unclear. In a study, urolithin A has reduced amyloid beta levels in mice models of Alzheimer's disease and decreased the levels of activated microglia and astrocytes. These mechanisms were hypothesized to suppress neuroinflammation and lead to the decrease of amyloid beta production.
