Hesperidin is a flavonoid that possesses a wide range of biological effects. It has been reported to improve microcirculation, reduce inflammation, improve symptoms of Rheumatoid Arthritis and lessen the swelling of the arms. An overlooked health benefit of hesperidin is its neuroprotective properties that may aid in the prevention of brain disorders, including autoimmune demyelinating disease, although the mechanisms of action need further investigation. Hesperidin has been shown to reduce pro-inflammatory cytokines, which interferes with the neuroinflammation in non-neuronal systems as well as the brain. Neuroinflammation plays a key role in the pathogenesis of Alzheimer's disease, Parkinson's disease, autism, and Parkinson's disease. Neuroinflammation is induced by a bacterial surface molecule lipopolysaccharide, which is associated with increased levels of oxidative stress and pro-inflammatory mediators, such as T-N-F alpha and interleukin 6. It has been postulated that the potential sources of these mediators are microglia and astrocytes. Hesperidin suppresses the biological activity of astrocytes, microglia, or both, therefore, reducing the production of pro-inflammatory mediators that contribute to neuroinflammation. In animal studies, hesperidin has been found beneficial in ameliorating the clinical abnormalities in the mice model of multiple sclerosis. Hesperidin decreases pro-inflammatory mediators, reverses oxidative damage, and alleviates lipid peroxidation and suppression of antioxidants, which are characteristics of multiple sclerosis.
