Low-dose naltrexone or LDN refers to a daily dose of 1 to 5 mg of te typical opioid addiction treatment dosage. Naltrexone hydrochloride is an F D A approved non peptide opioid antagonist approved in 1995 as a treatment of alcool addiction. LDN has been used as a treatment option for inflammatory disease such as Fibromyalgia, multiple sclerosis, and Chron's disease. Recent studies have reported that LDN could also be a potential treatment for COVID-19. SARS-COV-2 contains a surface protein, called receptor binding protein, with the host receptor, called, Angiotensin-Converting Enzyme 2 or ACE2. The interaction of the virus and the host cells leads to infection, which induces pro-inflammatory cytokine storm. A study publised in the Journal of Biomolecular Structure and Dynamics in 2020 has reported that L D N interferes with the binding of SARS-COV-2 spike protein to ACE2 and prevents hyperinflammatory cytokines release both macropage cells and Adipose tissue macrophage.
