Minoxidil is a vasodilator agent, indicated for severe refractory hypertension in the 1970's. However, its clinical application for hypertension has been discontinued due to its adverse effects. Some patients taking minoxidil have developed hypertrichosis. In 1987, a low dose topical form was developed to treat androgenetic lopecia. As a potent arteriolar vasodilator, it opens potassium channels located on the smooth muscles of the peripheral artery. It has been suggested that potassium channel activity may play a key role in cell proliferation at the early stage. It induces vascular endothelial growth factor expression increases vascularization, which may cause hair growth. Minoxidil also activates beta-catenin pathway, which delays the aging process.Moreover, it has been suggested to increase cellular DNA synthesis and enanced cell proliferation. Androgenetic alopecia is characterized by DNA damage leading to lower proliferation rate that results in follicular miniaturization.
