Rifampicin is an antibiotic drug that belongs to the class of rafamycins. Rifampicin was discovered in 1965 and has been used as a medication for the prevention and treatment of tuberculosis. Recently, rifampicin has been shown to increase lifespan in Caenorhabditis elegans by up to 60% through its ability to reduces advanced glycation end products or AGEs. AGEs are heterogeneous groups of molecules characterized by the combination of proteins or lipids with sugar in the bloodstream. The accumulation of AGEs in tissues has been proposed to alter the structure and function of proteins, which may contribute to the onset and progression of metabolic disorders and age-related diseases, as well as oxidative stress and inflammation associated with neurodegenerative diseases. The antiglycating activity of rifampicin is due to its ability to activate a transcription factor called DAF-16. It as been reported that the activation of DAF-16 leads to the upregulation of AGES detoxification system, which result in increased AGE disposal and a consequential increase in lifespan.
