The most important epigenetic mechanism is DNA methylation. It has gained research interest because it has been the easiest to study and it plays an essential role in many biological processes, including chromosome inactivation, gene suppression, and genomic imprinting. Moreover,abnormal DNA methylation has been established to be associated with cancer. So what happens during DNA methylation is that the enzyme DNA Methyl Transferase transfer s a methyl group onto a portion of cytosine to form 5-methylcytosine. Methylation inhibits the expression of a gene by recruiting proteins that are involved in gene repression. Changes in the DNA methylation have been associated with aging and aging-related diseases. Currently, it is an established biomarker of biological age and a predictor of mortality and susceptibility to diseases. A study has been conducted to characterize the epigenetic molecular basis of the human hippocampus in Alzheimer's disease. Hippocampal samples were collected from a cohort of pure Alzheimer's disease patients and controls. Successively, genome-wide DNA methylation levels were profiled using methylation arrays. As a result, Alzheimer's disease related differentially methylated positions or DMPs were identified in the Alzheimer's disease hippocampus. Alzheimer's disease related DMPs were preferentially involved neurodevelopmental and neurogenesis-related genes.
