Lipofuscin is a yellowish brown pigment found in different organs including nerve cells, retina, muscles, liver and ganglion cells. In cell culture models, oxidative stress has been shown to promote the accumulation of lipofuscin and lower the turnover of effective mitochondria, which may in turn diminish autophagocytotic capacity by interfering with recycling of cellular components. Autophagic activity is essential in mediating mechanisms that promote longevity, therefore, a decrease in this activity contributes to aging-associated diseases, including neurodegeneration and cancer. In the brain, lipofuscin is found in almost every type of neuron, but is prominent in areas involved in initiating, monitoring, and controlling movement. In an in vitro study, lipofuscin has been found to directly inhibit ubiquitin proteasome system in human neural cells. Ubiquitin proteasome system has the function to remove damaged proteins to permit the synthesis of new proteins. An upregulation of the components of ubiquitin proteasome system has been reported to extend lifespan. On the other hand, an impaired proteasome function due to lipofuscin has been associated with the pathogenesis of many neurodegenerative diseases.
